چکیده
Background: Osteonecrosis of the femoral head (ONFH) is a common disease caused by many trauma factors and un-trauma factors. Among those factors, steroid-induced osteonecrosis (SNFH) accounted for large proportion mainly concentrated in young people. SNFH has been reported as an irreversible associated with damage blood vessels loss balance bone homeostasis. Circulating endothelial progenitor cells (CEPCs), one part circulating (CECs), are immature precursor proliferative potential. The vascular confirmed studies, but changes CECs CEPCs peripheral patients have not studied yet. Objectives: objective study to explore number normal people then investigate whether EC-secreted exosomes (EC-exos) could prevent progression rat model its mechanism action. Methods: We collect 3 heathy detected levels Flow cytometer. TEM, NTA western blot was used characterize isolated EC-exos. Annexin V-FITC/PI double staining flow cytometric analysis were evaluate MC3T3-E1 apoptosis. CCK-8, scratching experiment transwell viability migration ability. Micro-CT morphological progress model. Results: Firstly, we found that decreased than Then our results indicated EC-exos improve migration, apoptosis osteoblasts under dexamethasone activating PI3K/AKT/Bcl-2 signal pathway vitro. Finally, revealed prevented SNFH. Conclusion: enhance cell ability play anti-apoptosis role against steroids-induced osteoblast pathway. Exosomes from Human Synovial-Derived Mesenchymal Stem Cells Prevent Glucocorticoid-Induced Osteonecrosis of the Femoral Head in the Rat
Osteonecrosis of the femoral head (ONFH) represents a debilitating complication following glucocorticoid (GC)-based therapy. Synovial-derived mesenchymal stem cells (SMSCs) can exert protective effect in the animal model of GC-induced ONFH by inducing cell proliferation and preventing cell apoptosis. Recent studies indicate the transplanted cells exert therapeutic effects primarily via a paracr...
متن کاملExosomes derived from human platelet-rich plasma prevent apoptosis induced by glucocorticoid-associated endoplasmic reticulum stress in rat osteonecrosis of the femoral head via the Akt/Bad/Bcl-2 signal pathway
An excess of glucocorticoids (GCs) is reported to be one of the most common causes of osteonecrosis of the femoral head (ONFH). In addition, GCs can induce bone cell apoptosis through modulating endoplasmic reticulum (ER) stress. Among the three main signal pathways in ER stress, the PERK (protein kinase RNA-like ER kinase)/CHOP (CCAAT-enhancer-binding protein homologous protein) pathway has be...
متن کاملExosomes Secreted from Human-Induced Pluripotent Stem Cell-Derived Mesenchymal Stem Cells Prevent Osteonecrosis of the Femoral Head by Promoting Angiogenesis
Background: Local ischemia is the main pathological performance in osteonecrosis of the femoral head (ONFH). There is currently no effective therapy to promote angiogenesis in the femoral head. Recent studies revealed that exosomes secreted by induced pluripotent stem cell-derived mesenchymal stem cells (iPS-MSC-Exos) have great therapeutic potential in ischemic tissues, but whether they could ...
متن کاملHigh Serum Alpha-2-Macroglobulin Level in Patients with Osteonecrosis of the Femoral Head
Background: Diagnosis of osteonecrosis of the femoral head (ONFH) is complicated due to the lack of reliable serum biomarkers. Up-regulation of alpha-2-macroglobulin (A2M) gene has been reported in glucocorticoid-induced ANFH rat model. This study aimed to investigate whether the serum level of alpha-2-macroglobulin (A2M) can be used for ONFH diagnosis. Methods: Serum protein capillary elect...
